This book examines the role of circadian rhythms in aging, an emerging area of biology. Although implicated in aging and longevity for over forty years, the richness of the ways in which the circadian system impacts aging has become evident only more recently. The circadian system consists of a central pacemaker and a multitude of peripheral clocks, located in most cells throughout the organism. These clocks keep metabolic, physiological, and behavioral patterns in tune with the twenty-four hour day/night cycle and with each other. Disruptions of the circadian system, such as the presence of light at night, can have profound pathological implications. The core circadian oscillator consists of a multicomponent transcriptional feedback loop that regulates the periodicity of expression of some ten percent of the genes in the human genome. This easily explains the broad influence of the circadian system.

The chapters in this volume fall into four sections that follow a brief introduction. They are written by experts performing the research in this field. The first section/chapter surveys the operation of the aging circadian system with a focus on melatonin signaling. In the second section, this theme is amplified through a discussion of the aging lung, bone, and gastrointestinal system. A section composed of eight chapters explores in detail cellular and molecular mechanisms associated with circadian system aging, in a range of experimental models. The studies reviewed include genetic, epigenetic, molecular, cell biological, metabolic, and physiologic approaches. The final section details the effects of sleep disruption on mortality risk in older adults and the effects of physical activity on circadian rhythms in the elderly, adding to the earlier discussion of the potential of chronotherapeutics.

About the Author

S. Michal Jazwinski, Ph.D. is the John W. Deming, M.D. Regents Chair in Aging, Professor of Medicine, and Director of the Tulane Center for Aging at Tulane University. His research on various aspects of aging has been funded by the National Institute on Aging (NIH) and private foundations, and it spans over thirty years. It includes genetic studies in model organisms and multidisciplinary studies in humans.

Victoria P. Belancio, Ph.D. is an Associate Professor of Structural and Cellular Biology at Tulane University. Her research on retrotransposon-associated genomic instability has been funded by the National Institute on Aging (NIH) and private foundations. It includes projects investigating how circadian disruption impacts DNA damage induced by retroelements and whether this damage influences mammalian aging.

Steven M. Hill, Ph.D. is the Edmond and Lily Safra Chair for Breast Cancer Research, Professor of Structural & Cellular Biology, and Director of the Tulane Center for Circadian Biology at Tulane University. His research on various aspects of circadian biology and breast cancer has been funded by the National Cancer Institute (NIH), the Army Department of Defense Breast Cancer Program, and private foundations, and spans over 25 years. His recent research efforts have focused on the impact of circadian/melatonin disruption by light at night on human breast cancer progression, metastasis, and drug resistance.

Astrovirus Research: Essential Ideas, Everyday Impacts, Future Directions

Astroviruses were first identified in the feces of children in 1975. Since then, they have been found in ~ 3 to 20% of children with diarrhea. Given that serological studies have demonstrated that up to 90% of children have been exposed to at least one strain of astrovirus by age 9, the prevalence of infection may be much higher. Supporting this are studies demonstrating that astroviruses can also be isolated in a subset of asymptomatic individuals, suggesting that a proportion of infected individuals shed the virus asymptomatically or for some time after the resolution of other symptoms of infection. Asymptomatic carriers may be a major reservoir for astroviruses in the environment and could contribute to dissemination of the virus. Astroviruses are extremely stable in the environment and can be transmitted nosocomially, directly from infected individuals and potentially animals, and through contaminated food and water. Although typically an acute disease, astrovirus infection in premature infants may be associated with the development of necrotizing enterocolitis and in new-onset celiac disease in children. Immunocompromised children are even more susceptible often developing persistent infections that lead to wasting or even systemic infections associated with fatal encephalitis. In spite of its importance, little is known about astrovirus pathogenesis, molecular biology, epidemiology, or cell biology. The goal of this book is to provide the latest and most up-to-date information on this medically important and rapidly evolving group of viruses. It will include sections on the history of astroviruses and their disease in humans; information on viral replication and immune responses; new information on how astroviruses induce disease including the expression of a viral enterotoxin regulating intestinal epithelial cell tight junctions, the isolation and identification of new astrovirus genotypes in mammals including humans, and astroviruses of veterinary importance. Finally, the book will also introduce the complexity of astrovirus epidemiology and potential as an important new zoonotic disease, and its role in food-borne disease. This will be the first book of its kind and will be of great interest to microbiologists, virologists, infectious disease specialists, immunologists, pediatricians, public health and food safety experts, veterinarians, poultry industry specialists, and researchers and clinicians interested in enteritis.

Siba P. Dubey, MBBS, MS

Siba Dubey obtained his MBBS degree from Calcutta University in 1982. He trained in Otolaryngology – Head and Neck Surgery (ORLHNS) at the Postgraduate Institute of Medical Education and Research (PGIMER), Chandigarh, India. He completed his thesis on “Computed tomography (CT) in Ear diseases – Correlation between CT and surgical findings” and passed Master of Surgery (MS) in ORLHNS in December, 1986. Subsequently, he underwent advanced training in ORLHNS in India and Austria. He came to Papua New Guinea (PNG) in November 1994 to take up the position of Chief Ear, Nose and Throat (ENT) Surgeon. The specialty of ORLHNS grew bigger from one to multiple centres across PNG with the help of locally trained ENT specialists. He became Professor at the School of Medicine and Health Sciences, University of Papua New Guinea in 2002. Since then, he has been involved in the teaching and training of Undergraduate and Postgraduate students of PNG and other Pacific Island countries.

He has performed research on clinical and operative aspects, malignancies of head and neck, and tropical diseases, namely, angiofibroma, scleroma etc. These works are published in international journals and in books.

Bernhard Schick, MD

After studying medicine at the Philipps-University in Marburg, Germany from 1987 to 1993 he completed his doctor thesis in 1993. Entitled “Enzymatic analysis of myosin-subfragment-1 induced actin polymerisation” the thesis was honoured by the note “summa cum laude”. ENT specialization took place under the supervision of Prof. Dr. Dr. W. Draf (Fulda-Hospital, Germany) from 1994 ton 1998. Specialization for plastic head and neck surgery was successfully achieved under the supervision of Prof. Dr. Dr. W. Draf (Fulda Hospital, Germany) from 1998 to 2000. As the representative of the Head at the ENT-Department at the Saarland University (Prof. Dr. P.K. Plinkert) from 2000 to 2003 habilitation was successfully finished in 2003, presenting new findings of inner ear mechanics analysing VASP expression in the inner ear with the chance to analyse hearing in VASP-/- mice by detailed electrophysiological examinations. He has been Head of the ENT-Department at the Saarland University since June 2008, and is now Medical Director of Uniklinikum Saarland and CEO.

During a 20 year period extensive studies have been performed on juvenile angiofibromas, from genetic and molecular studies to clinical evaluations including radiological tumor presentation, surgical strategies and development of an etiological model describing tumor genesis.
